Abstract
Understanding and predicting signaling pathways is a top research priority by pharmaceutical industry and academia. This because behavior, response to stress, regulation and function of human cells and organs they form is orchestrated by signal transduction. Normal and healthy function of cells and organs is ensured by cell-cell connectivity and by a much more complex dynamic of biochemical intracellular signals transmission through a network of cooperative interactions, which include protein-protein gene-gene, DNA-proteins and other molecules interactions. Here the advances in pathways analysis and multiple challenges understanding how genetic and epigenetic alterations modify transcriptional activity, chromatin modeling and expression of multiple interacting genes leading to deregulation of signaling networks and diseases are discussed.
