Overexpression of Erbb2 in Breast Cancer: Associated Signal Transduction and Termination Events

ABSTRACT

Human epidermal growth factor receptor 2 (HER2 or ErbB2), is a member of subclass I of the superfamily of receptor tyrosine kinases. It has a vital role in breast cancer. Overexpression or amplification of ErbB2 is observed in many breast cancer patients, in whom, normal cells turn out to be cancerous, and lead to poor prognosis. ErbB2 receptor protein is involved in the activation of several signal transduction events: Ras-MAPK, IP3/DAG, PI3-K and JAK/STAT pathways, respectively. In this review, the role of ErbB2 in breast cancer is discussed and the current status of the related ErbB2 receptor pathways induced by overexpression and signal termination events are presented.
